The band started off as a hobby group in Huntsville. Claude McKnight, the founder of the group, which was initially called the The Gentlemen's Estate Quartet, began to audition other students for an a capella quartet at Oakwood University. For the next several years the group performed on campus and at local churches where they began to gain much attention. The quartet started to welcome additional members and eventually evolved into the sextet that we know today. In 1987 the group was signed to the renowned record label, Warner Brothers. It was then that the name Take 6 was adopted by the members.

Take 6 specialized in singing a capella music, a style in which group or solo singing takes place with little to no instrumental sound. Starting out as mainly gospel, their music style evolved over time to include elements of jazz and R&B. This further helped them to garner more attention nationwide. Fans are especially fond of their harmony-rich sound which is different from that of any other contemporary music group.

The music of Take 6 has been featured in numerous films such as Boyz n the Hood and Spike Lee's Do the Right Thing. Apart from these it is featured in the television series Martin. With performances at the Oscars, the Grammys and the White House, it is no surprise that they have such a large fan base all over the country.

Besides playing at impressive events for notable public figures, Take 6 has appeared on tour with many other artists. These include Stevie Wonder, Ray Charles, Whitney Houston, CeCe Winans, Queen Latifah and many other famous artists. The group quickly rose to fame and dominated the music charts in the US. Their first album release placed third on the US Gospel chart and eighth on the US Jazz chart. Their next album, titled So Much 2 Say, again took the eighth spot on the US Gospel chart and second on the US Jazz chart.

In 2008, Take 6 released an album titled The Standard which placed second and sixth on the US Jazz and US Gospel charts respectively. Their realistic sound and peaceful harmonies quickly become favorites around the country.
